The first task is to make F4808N measurable in human muscle.
F4808N has no direct public functional measurement. The project needs a qualified human-muscle model and a repeatable result.
No iPSC line, muscle model, or functional result exists yet.

The biological question
What must the model resolve?
RYR1 helps skeletal muscle release calcium for contraction.
Different RYR1 variants can cause different effects.
The project does not yet know the F4808N mechanism.
A reliable phenotype can identify the next valid treatment test. A valid negative result can prevent the wrong work.

What must the lab build?
Each work unit must give a checkable output for the next unit.

Generate the model
Generate independent F4808N iPSC clones and corrected-isogenic controls. Release only clones that pass the agreed quality checks.

Make skeletal muscle
Differentiate independent clones into skeletal muscle. Confirm a stable muscle state and sufficient RyR1 expression.
- 03
Measure the phenotype
Measure allele-specific RNA, total RyR1 protein, resting calcium, stored calcium, stimulated calcium release, and stimulated force.

Prove repeatability
Repeat each important measurement across independent clones and test runs. Lock the methods, quality rules, and analysis before the final comparison.

Which outputs must connect?
Each output tests a different part of the biological question.
The planned measurements are allele-specific expression, total RyR1 abundance, baseline calcium state, calcium available for release, response after stimulation, and functional muscle output.

The value is not one attractive trace. The value is a linked data package that another laboratory can inspect and repeat.

A usable result
- The effect separates F4808N muscle from a matched control.
- The effect repeats across independent clones and test runs.
- The effect moves toward the control state after correction.
- Another laboratory can repeat the locked test.
A reason to revise or stop
- The effect changes with the clone, batch, or analysis method.
- The model does not produce a valid RYR1 measurement.
- The effect does not survive one defined optimization cycle.
